Background staining can be classified into two categories: one is immunological in origin, that is, it is due to cross-reactivity of the antibodies, and the other is methodological in origin related to the reagent itself. Immunochemical reagents theoretically will not react; this is found to be untrue as electrostatic and hydrophobic interactions of immunochemical reagents with fixed or unfixed tissues occur frequently, requiring bovine serum albumin (BSA), gelatin, or normal serum (blocking solution) for saturating the nonspecific binding sites. Nonspecific staining peculiar to the second antibody method can be recognized by staining the tissues only with the probe-labeled second antibody. Another source of nonspecific staining in the immunoperoxidase method could be the endogeneous peroxidase of the tissues." @default.
