- W336870295 abstract "The biosynthesis of the benzoisochromane-quinone antibiotic nanaomycin (NNM) and the 16-membered macrolide antibiotic spiramycin was studied by means of a bioconversion method using cerulenin, an inhibitor of fatty acid and “polyketide” biosyntheses, and then some enzymes involved in the biosynthetic pathway thus proposed were characterized using purified or partially purified preparations. In the biosynthesis of NNMs by Streptomyces rosa var. notoensis, NNM-D was considered to be the first component produced from “polyketide”, and the biosynthetic sequence was suggested to be: NNM-D → NNM-A → NNM-E → NNM-B. A flavoprotein which catalyzed the formation of NNM-A from NNM-D was isolated from the producer and characterized. In the biosynthesis of spiramycin by S. ambofaciens, the biosynthetic sequence was proposed to be: platenolide I → forocidin I → neospiramycin I → spiramycin I → spiramycin II. Spiramycin I 3-hydroxyl acylase which catalyzed the formation of spiramycin II (and III) from spiramycin I was partially purified. From the substrate specificity the acylation was supported to occur in the last step of the biosynthetic pathway.
