- W341680312 abstract "Amyloidosis is a heterogeneous group of disorders caused by extracellular deposition of a characteristic proteinaceous material, the amyloid substance, in various organs and tissues (Glenner 1980; Husby and Sletten 1986a, b). The pathophysiological consequences of amyloid deposition depend on its tissue and organ distribution, and the resulting symptoms and signs are partly caused by functional disturbances due to displacement of normal tissue structures, and partly by organ enlargement (Husby 1983). Benditt and Eriksen (1964) suggested for the first time that amyloidosis was heterogeneous also with respect to the nature of the amyloid deposits. It was, however, difficult to isolate the fibrils from affected tissues because of their very low solubility in physiological conditions. This property of amyloid was thought to be responsible for the irreversible and often progressive course of the disease, which in many cases leads to death within months or years of diagnosis (Husby 1983, 1985). Important progress was made when Pras et al. (1968) published their water extraction method for the purification of amyloid fibrils. They confirmed suggestions made more than a century earlier, that amyloid was principally of protein nature, although Virchow in 1854 had created the term amyloid (starch-like) because the deposits stained with iodine in a manner similar to that of starch or cellulose." @default.
