• W345367128 abstract "Publisher SummaryThis chapter discusses studies on the localization of dopaminergic and cholinergic receptors in the carotid body. Tissues from two groups of rabbits and cats were used in this study. In one group, the carotid sinus nerves were bilaterally resected 12–15 days prior to experimentation; the other group included normal, unoperated animals. Carotid bodies were removed for binding assay from pentobarbital-anesthetized animals and cleaned of surrounding connective tissue in a Lucite chamber filled with O2-equilibrated, modified-Tyrode's solution. Tissues were incubated for 20–30 min in a waterbath-shaker at 37°C. Total and nonspecific binding for each of the ligands were determined in the absence and presence, respectively, of agents that competitively protected the receptor sites. The results show that 3H-spiroperidol binding in rabbit carotid body was concentration-dependent. Specific binding was saturable at spiroperidol concentrations above 0.38 nM, and the time course of binding at this concentration reached a plateau after 20 min of incubation. The absolute amount of nonspecific spiroperidol binding in both normal and denervated carotid bodies is comparable to that reported for human caudate nucleus." @default.
