Para identificar el papel regulador del operon narC(GHJI) y el AMPc se const ruyeron cepas isogenicas solo presentando la fusion genomica ( moa::lac )25, o con mutaciones adicionales. La expresion de la fusion fue evaluada determinando la biosintesis de la Gal , codificada por el gen lacZ fusionado al promotor Pmoa, en cultivos crecidos con medio sintetico durante una noche. La presencia de una mutacion cya disminuyo parcialmente los niveles de actividad especifica Gal. por el contrario, la mutacion narC-8 resulto en la incapacidad de sintetizar Gal , en cepas cya+ y cya. Este efecto no fue eliminado por crecimiento bajo condiciones de induccion del operon narC. El AMPc exogeno no abolio los efectos de la glucosa ni de la mutacion narC-8. Los resultados apoyan la participacion de mas de un control positivo que actuaria regulando la expresion transcripcional del operon moa, y en consecuencia afectarian la biosintesis del MoCo. EnglishIn Escherichia coli K12, catabolite repression and nar region globally regulate operons induced by anaerobiosis. In the nitrate anaerobic respiration, previous studies evidenced to the glucose as regulator of the moa operon, which participates in the molybdocofactor biosynthesis, MoCo, of the nitrate reductase (NRA). To identify the regulator role of the narC(GHJI) operon and the cAMP, were constructed isogenic strains carring either the ( moa::lac )25 genomic fusion, or plus additional mutations. The fusion expression was evaluated determining the Bgal biosynthesis, encodied by the gen lacZ united to the Pmoa promoter, in culture overnight growing with minimal medium. The presence of a cya mutation partially decreased the specific Gal activity levels. In contrast, a narC-8 resulted in the inability to synthesize Gal, in both cya+ and cya strains. This effect was not eliminated by growin g under conditions that induce the narC operon. The exogenous cAMP either did not abolish the glucose nor narC-8 mutation effects. The results support the participation of more than one positive control that should act on the transcriptional expression of the moa operon, in consequence it should affect the MoCo biosynthesis." @default.
