- W35596529 abstract "This chapter focuses on human neuronal cell lines as in vitro models. The chapter describes the general development of a culture strategy and the various parameters, which may be tried in order to establish optimal cell growth and differentiation. Several new techniques have emerged that may be used to generate cell lines. A particularly successful strategy has utilized oncogene containing retroviruses. This requires a relatively high density of dividing cells. Viral oncogenes that are able to transform cells and, thereby immortalize them, are infected into recipient cells and during the process of DNA replication, become inserted into the genome and are therefore, stably transmitted to progeny. Another method that has been used to generate neuronal cell lines is somatic cell hybrids. A continuous cell line may be fused with a desired cell, such as a specific neuronal cell, and this may generate somatic cell fusions that are capable of continuous propagation. A problem with some somatic cell hybrids is that the chromosome content is unstable and cells may subsequently shed chromosomes, altering their phenotype. This chapter discusses in detail the selection of tissues and preparation of cells for plating. The chapter also explains maintenance of cell cultures and differentiation of neuronal cell lines. Characterization of neuronal cell lines is also discussed in detail in this chapter." @default.
