- W366828395 abstract "The time course of changes in endogenous phospholipids, induced by ionophore A23187, collagen and thrombin, was compared to arachidonate (AA) oxygenation and changes in chlortetracycline (CTC) fluorescence, an indicator of membrane-bound calcium. The data indicate that AA oxygenation, and thus phospholipase A2 activity, preceeds changes in phosphatidylinositol (PI) and phosphatidic acid (PA), and suggest that thrombin-induced calcium release from membrane sites may be responsible for activation of this enzyme. In contrast to thrombin, release of membrane-bound calcium upon ionomycin addition was minimal, suggesting that ionophores release calcium from another site. A difference in pattern of phospholipid hydrolyses with A23187, notably a slight lag period and a lack of PA generation, is consistent with this conclusion. In contrast, the initial breakdown of endogenous phosphatidylinositol 4, 5-bisphosphate (PIP2) upon platelet stimulation paralleled changes in CTC-fluorescence, suggesting a close link between PIP2 breakdown and release of membrane calcium. Calculations indicate that cytoplasmic free (C{{a}^{{{{2}^{ + }}}}}) in stimulated human platelets could be derived from calcium bound to stimulus-sensitive PIP2." @default.
