SUCLA2 -/- eger ezidaig nem szuletett (lehet, hogy perigasztralis elhalalozassal jar); v) az MDH2 transzgenikus egerek (b varians) nem mutattak csokkent MDH aktivitast, igy nem kereszteztuk őket egyik fenti torzzsel sem; vi) kiserleteink soran arra a kovetkeztetesre jutottunk, hogy a legzesi lanc cytochrome b5 reduktaza (cyb5r) az altalunk tapasztalt jelenseg fontos mediatora, es a PI elnyert egy kisebb palyazatot mely fedezi a cyb5r 2-es izoformara kiutott egerkolonia beszerzeset az EUCOMM-tol. Jelenleg ezek az egerek szaporitas alatt vannak az EOK SPF allathazaban. Kiserleteink ramutatnak a mitokondrialis diaforazok fontossagara a matrix NADH/NAD+ arany fenntartasaban, mely temat a jovőben aktivan vizsgalni fogjuk. | The progress on grant NNF2 85658 is as follows: i) all experiments on isolated brain and liver mitochondria, isolated nerve terminals, and cultured neurons and astrocytes from WT, DLD+/-, DLST+/- mice have been completed; ii) most experiments on DLD+/- DLST+/- double KO mice have been completed on isolated brain and liver mitochondria, and isolated nerve terminals; iii) all experiments on DLD+/- cypD-/- double KO mice have been completed in isolated brain and liver mitochondria. No experiments have been performed yet on DLST+/- cypD-/- double KO mice; iv) the SUCLA2 mice have finally been generated, as of June 1st we have 4 male +/- and 4 female +/- mice. No -/- SUCLA2 mice have been born yet (perhaps this is associated with perigastrulation lethality); v) the MDH2 transgenic mice (b variant) did not exhibit a decreased activity in MDH, therefore they were not crossed with any of the other transgenic mice; vi) in the course of our experiments we deduced that an important component mediating the phenomena that we observed is the cytochrome b5 reductase (cyb5r) of the respiratory chain, and the PI won a small grant (waving the costs for obtaining a transgenic mouse colony) for obtaining cyb5r isoform 2 knock-out mice from EUCOMM. The mice are currently breeding in our SPF facility of the EOK. Our experiments point out the importance of mitochondrial diaphorases participating in the maintenance of matrix NADH/NAD pools, and this will be actively pursued in the future." @default.
