- W375087149 abstract "Fluorescent probes are valuable molecular tools in chemical biology, and fluorogenic probes are particularly useful because they can suppress noise associated with background emission. Several tag/probe pairs have been developed for the fluorescent imaging of proteins. Previously, we developed a new tag/probe pair with fluorogenic activity using the leucine zipper assembly, which was designated as the ZIP tag/probe [1,2]. A probe peptide was designed as an alpha-helix peptide, which had an solvatochromic fluorescent dye, 4nitrobenzo-2-oxa-1,3-diazole (NBD). Tag peptides was designed as an antiparallel 2 alphahelix peptide, and the tag and probe form the 3 alpha-helical bundle structure of the leucine zipper. The hydrophilic environment surrounding NBD of the probe peptide is drastically changed into the hydrophobic environment through the formation of a 3 alpha-helical leucine zipper structure between the tag and the probe peptides as a result of which the bright green fluorescence of the NBD dye is induced. In this study, 7-diethylaminocoumarin-3-carboxylic acid (DEAC) was used in the development of another ZIP tag/probe pair with switchable blue fluorescence [3]. Fluorescence switching of DEAC from the dark to bright blue was achieved through a formation of a 3 alpha-helical bundle structure. The DEAC unit in the leucine zipper assembly showed 50 times higher fluorescence than that of DEAC unit alone. The stable 3 alpha-helical bundle structure was essential for the great enhancement of the DEAC fluorescence. The fluorescence enhancement of the DEAC-probe peptide by the binding to the tag peptides can be easily detected by the naked eye.
