- W37653073 abstract "Cell walls have been described since the 17th century. This outer layer, surrounding the plasmalemma, was long considered as a passive, inert, plant skeleton, an intercellular cement which directed the shape of cells and consequently of the whole plant. Cell walls account for 20 to 60% of tissue dry matter. This insoluble material has been used by mankind since the beginning of time and its chemical composition has been thoroughly investigated throughout the 20th century. However, the wall’s physiological functions were recognized only relatively recently. Cell walls may be considered from at least three different view points. (1) As a cell compartment between the external medium and the protoplasm, this barrier will affect absorption and exorption processes. (2) As a rigid envelope, its mechanical properties will limit the extension potential of the cell. (3) As the most external cellular layer, it is involved in recognition processes: for instance, some cell-wall polysaccharides might act as effectors in recognition systems between host and pathogens (Albersheim et al. 1980). Recently, knowledge of wall functions has greatly expanded and many experiments have been carried out on isolated cell walls. The purpose of this chapter is to answer two questions: first, how can pure isolated cell walls be obtained; second, what aspects can be studied with these isolated cell walls. We shall systematically consider the improved methods used for isolating cell walls and then the approaches adopted to investigate their composition, structure, and properties.
