- W37954122 abstract "200 Objectives Both 68Ga-DOTATATE and 68Ga-DOTATOC are used for PET/CT evaluation of neuroendocrine tumours (NETs). The aim of the present work was to evaluate the kinetics of both tracers in NETs and to validate the use of standardized uptake values (SUV) as a measure of somatostatin receptor expression. Methods Ten patients with NETs underwent 45 min dynamic PET/CT scans after injection of 90 MBq of either 68Ga-DOTATATE or 68Ga-DOTATOC, followed by whole-body scans at 1, 2 and 3 h p.i.. PET/CT was repeated the following day with the other tracer. Volumes of interest (VOIs) were drawn on 15-45 min sum images at 70% isocontours over all clearly identifiable lesions with diameters > 2 cm, and projected onto all frames of the dynamic scan. Arterial input functions were based on a descending aorta VOI and venous blood samples. Data were analysed using single tissue (2k), two-tissue irreversible (3k) and reversible (4k) compartment models as well as the Patlak method. Distribution volume, net uptake rate (Ki) or binding potential as estimated by 2k, 3k and 4k models, respectively, was assumed to reflect somatostatin receptor density. SUV was based on 1 h whole-body images. Results For both tracers, only the 3k model described the data well. Ki values based on the Patlak method correlated well with those based on non-linear regression of the 3k model (R2 = 0.95 and 0.96 for 68Ga-DOTATATE and 68Ga-DOTATOC, respectively). For both tracers, correlation between SUV and Ki was moderate (R2 = 0.78 and 0.79) and SUV appeared to saturate for Ki > 0.2 ml/cm3/min. Conclusions Kinetics of both 68Ga-DOTATATE and 68Ga-DOTATOC are irreversible during the first hours after injection. For both traces, correlation between SUV and Ki was moderate and SUV may not reflect somatostatin receptor density at higher values." @default.
