- W407214 abstract "This chapter describes the immunostimulating complex, referred to as an “iscom” or ISCOMTM vaccine, as a novel structure for antigenic presentation of membrane proteins from enveloped viruses with potent immunomodulatory capability. Since this discovery, many ISCOMTM vaccines have been tested in animal models showing the induction of both humoral and cellular immune responses. ISCOMATRIXTM adjuvant is essentially the same structure as the ISCOMTM vaccine but without the incorporated antigen. Antigens can be formulated with the ISCOMATRIXTM adjuvant to produce ISCOMATRIXTM vaccines providing the same antigen presentation and immunomodulatory properties as ISCOMTM vaccines but with much broader application. Various ISCOMATRIXTM vaccines have been tested in animal models and more recently in human clinical trials. All results to date indicate that ISCOMATRIXTM vaccines are safe and well tolerated in humans with no vaccine-related serious adverse events or laboratory abnormalities. ISCOMATRIXTM vaccines are capable of inducing strong humoral responses with increases in the magnitude, speed, and longevity of the responses, as well as the capacity for antigen dose reduction when compared to other adjuvants such as aluminum. The ability of the ISCOMATRIXTM adjuvant to induce these broad immune responses is due to the fact that it is both an effective antigen delivery vehicle, which can access both the MHC I and MHC II pathways, and a potent immunomodulator of both the innate and adaptive immune systems. Overall, the properties of ISCOMATRIXTM vaccines make them viable candidates for the further development and registration of prophylactic and therapeutic human vaccines." @default.
