- W413405110 abstract "Objective: To study the effects of dark, lighting and behavior limitation on ischemic cardiac myocyte apoptosis and expression of Bcl-2, Bax and Caspase-3 in rats with myocardial infarction(AMI).Methods: Forty Wistar rats were randomly divided into five groups with 8 in each group: sham-operation group(group A), AMI group(group B), dark group(group C), lighting group(group D), and behavior limitation group(group E).Apoptosis of cardiac myocyte was detected with TUNEL method, and the expression of Bcl-2, Bax and caspase-3 were examined with immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR).Results:(1) Apoptosis indexes of groups B, C, D and E were higher than those of group A(P<0.01).Apoptosis indexes of groups D and E were higher than those of group B(P<0.05).Apoptosis indexes of groups D and E were higher than those of group C(P<0.05).(2) The expression of Bcl-2, Bax and Caspase-3 protein in groups B, C, D and E were higher than those of group A(P<0.01).The expression of Bax and Caspase-3 protein of groups D and E were higher than those of group B(P<0.05).The expression of Bax and Caspase-3 protein of groups D and E were higher than those of group C(P<0.05). (3) The expression of Bcl-2, Bax and Caspase-3 mRNA in groups B, C, D and E were higher than those of group A(P<0.01).The expression of Bax and Caspase-3 mRNA of groups D and E were higher than those of group B(P<0.01).The expression of Bax and Caspase-3 mRNA of groups D and E were higher than those of group C(P<0.01).Conclusions: Lingting, behavior restriction could increase ischemic cardiac myocyte apoptosis in rats. The mechanisms may be associated with up-regulating the expression of Bax and Caspase-3.Dark environment does not increase cardiac myocyte apoptosis in rats." @default.
