Matrix-assisted laser desorption mass spectroscopy (MALDI–MS) is a powerful soft ionization tool for the analysis of a wide range of organic and biological molecules using organic matrices. Nanoparticles and functionalized nanoparticles have been successfully applied as probes to enrichment of preconcentrated peptides, phosphopeptides from digested known proteins, or real-world samples. Nevertheless, miniaturization protocols based on metal nanoparticles are promising and effective for low detection limits from complex solutions of less than 1–2 μl sample by MALDI–TOF–MS analytical techniques. This chapter explores various metal nanoparticles, oxide nanoparticles, and their functionality as solid-phase extracting probes and various techniques such as single-drop microextraction (SDME) and liquid–liquid microextraction (LLME), on-particles proteins digestion, and enrichment of phosphopeptides and peptides under microwave environment. Nanoparticles with high affinity and bonding to bacterial cell wall proteins and antibacterial activities are covered. Nanoparticle arrays and functionalization-based self-assembled monolayers (SAMs) on biochips, which are hydrophobic/hydrophilic in nature, can be used to enrich bacteria for direct MALDI–TOF–MS analysis." @default.
