- W4200417076 abstract "Gestational diabetes (GDM) is associated with adverse pregnancy outcomes. Metabolic dysfunction with diabetes can alter immune responses. With GDM, changes in the placental immune system are thought to drive pregnancy complications. The objective of this study as to evaluate placental pro and anti-inflammatory gene expression in pregnancies complicated by GDM. Intervillous biopsies from GDM (n=8) and uncomplicated pregnancies (n=10) were acquired from term cesarean sections. An aggregated analysis of over 1400 genes related to immunity and cellular remodeling was conducted using two NanoString nCounter panels; the Myeloid Innate Immunity V2 and Fibrosis V2 panels. Gene expression analysis and the corresponding volcano plot were generated using NanoStrings nSolver software. Tissue sections were staining for CD163 with a Leica BondMax and >1mm2 regions of intervillous and decidua were scanned at 40x using a fully motorized Leica LMD6 microscope. Within scans, macrophages were quantified by counting CD163 immunostained cells and normalizing to tissue area using Leica’s LAS-X analysis application. Gene expression reveals a host of genes altered with GDM (Figure 1). In particular, genes coding for class II HLA’s (DQA1 and DQB1) were downregulated whereas pro-inflammatory genes, notably CCR7, IL15RA, and IL5RA were upregulated. No difference with GDM compared to normal were observed in the number of CD163 cells, though overall there were less in the decidua vs. the intervillous (Figure 2). We successfully identified differences in placental pro and anti-inflammatory gene expression with GDM. Type II HLA’s are indicative of anti-inflammatory placental macrophages. Pro-inflammatory macrophages express CCR7, IL15RA, and IL5RA. Since no difference in the number of CD163 cells was observed, gene expression results suggest that the placental macrophages shift from anti to pro-inflammatory within GDM. The shift of placental macrophages to a pro-inflammatory state requires further investigation.View Large Image Figure ViewerDownload Hi-res image Download (PPT)" @default.
