Synthetic biologists attempt to decipher design principles of naturally occurring genetic circuits to enable construction of complex multigene synthetic circuits. Mathematical modeling of genetic networks and their computational design are being successfully employed to guide construction of de novo genetic circuits. This chapter provides an introduction to design principles of synthetic genetic circuits and the approaches employed to construct circuits. Mathematical frameworks used to model synthetic genetic circuits are described, along with examples. A number of computational tools have been developed for design, modeling, and analysis of synthetic genetic circuits, and an overview of a few prominent ones is provided. The chapter concludes with a discussion of challenges in circuit design." @default.
