Learn more.Copy URL Share a linkShare onFacebookTwitterLinked InRedditWechat Abstract Decrease in LV mass and sarcopenia is a known characteristic of doxorubicin (Dox) cardiac toxicity. Postulated mechanisms include cell loss as well as disrupted sarcomere synthesis. We hypothesized that disruption of cardiac ankyrin repeat protein (CARP) plays a mechanistic role in the Dox-induced sarcopenia. Results: Short interfering RNA (siRNA) developed against CARP suppressed expression by over 75% at 24 hours in adult rat ventricular myocytes (ARVM). CARP protein levels were decreased by 12 fold at 24 hours when measured by western blot, similar to Dox (15 fold suppression at 24 hours). Myocytes treated with CARP siRNA were assessed for sarcomere integrity by immunostaining (myomesin, CARP, actin). Base line CARP staining showed two populations of CARP: a I-band population and a nuclear fraction. A third population of CARP was detected in freshly isolated myocytes as a central core running along the long axis of the myocyte. Immunostaining after treatment with CARP siRNA led to a 40% decrease in CARP content, a 51% decrease in myomesin and a 34% decrease of actin by 72 hours. Western blot of actin confirmed a decrease in total actin content in CARP suppressed cells. Conclusion: Suppression of CARP expression in ARVMs in vitro is sufficient to induce a sarcopenia similar to that induced by Dox, suggesting CARP is an important regulator of myocyte integrity.
