- W4210921212 abstract "Infectious diseases remain a major problem throughout the world. It is clear, at least for viruses, that prevention is our best defense. Most successful vaccines (e.g., smallpox, rubella, Sabin poliomyelitis, measles, mumps, yellow fever) have consisted of live attenuated viruses. In general, they provide long-lasting immunity, high potency, and economy of manufacture and delivery. A few vaccines (e.g., Salk poliomyelitis, influenza, rabies) are killed viruses. One vaccine (hepatitis B) consists of a purified subunit protein. Developments in recombinant DNA technology have opened new possibilities for vaccine production. Genetic engineering can be used to produce subunit proteins, attenuate viruses by gene deletion or modification, or construct live recombinant vectors. As a potential vector, vaccinia virus has impressive credentials, as it was responsible for the eradication of smallpox. Studies suggest that vaccinia virus can be used for expression of genes from other microorganisms and that such recombinants retain infectivity, induce humoral and cell-mediated immunity, and provide significant disease protection to experimental animals." @default.
