The success in obtaining effective enzyme–antibody conjugates seems to depend mainly on the purity and affinity of antibody, on the enzyme preparations used, and on the cross-linking reagent employed. To couple an enzyme with a protein, a bi- or multifunctional reagent is needed. This reagent should be specially selected when an enzyme has to be coupled to an antigen or to an antibody with the view to obtaining a protein–enzyme complex that retains a significant part of its immunological and enzymic activities. In early experiments, enzyme-labeled antibody prepared with a variety of coupling agents and enzymes often failed to detect the antigens. Enzyme-labeled antibody has an advantage over fluorescein-and ferritin-labeled antibody in that the same preparation can be utilized both for light and electron microscope studies. It is perhaps too early for the use of immunoenzyme techniques for the ultrastructural localization of antigens to be considered routine methods, but their potential seems as great as the immunofluorescence method for optical microscopy." @default.
