Singleuse manufacturing has become prevalent for clinical development and increasingly also for the commercial-scale manufacturing of Monoclonal antibodies (mAbs). This chapter discusses each of these unit operations in further detail and aims to provide the reader with an understanding of the fundamental principles as well as practical considerations during process development. It describes how these unit operations come together in an industrial context to form the harvest and clarification process for mAb products. Tangential flow or cross-flow microfiltration competes with centrifugation as the method of choice for the harvest of therapeutic products from mammalian cell cultures. In terms of placement in a harvest and clarification scheme for mammalian cell culture broths, depth filters are typically placed after a centrifugation step. Flocculants have been used for many years to improve the filtration characteristics of fermentation broths." @default.
