He eloquently discussed a variety of anti-inflammatory and other beneficial effects of heme oxygenase-1 (HO-1) that have been described in other disease states. Surprisingly, despite the intense interest in HO-1 in other conditions such as vascular disease,2 investigation of the effects of this classical, yet fascinating, enzyme has been limited in the human liver. We are excited by the possibilities of using HO-1 and heme oxidation products for drug design and additional therapeutic strategies for patients with chronic hepatitis C virus infection. Consequently, the major point of our article was that HO-1 induction or overexpression leads to both inhibition of viral replication as well as reduced oxidative hepatic injury. We feel the stage is now set to test the effectiveness of drugs that induce HO-1 as their mechanism of action for virucidal and anti-inflammatory activities in vivo. Warren N.
