For its control, studies of TB vaccines are needed. Since BCG is the only vaccine against TB currently in use, studies addressing the protective role of BCG is urgent required. Methods: Cohorts of HIV-negative voluntaries have been enrolled: adult Healthy Donors (HD) and neonates Umbilical Vein (UV). Host mononuclear cells were purified directly from whole blood, then BCG Moreau, Pasteur and Danish strains were primarily used for in vitro monocyte infection at 48h, and Annexin V-FITC and PI were employed for apoptosis detection by a flow cytometry approach. The bacilli viability was promptly assessed by an immunofluorescence kit (LIVE/DEAD® BacLight). The supernatant of the cultures were further used for cytokine detection assays. The P levels were setup at < 0.05. Results: Here, the BCG Moreau induced higher apoptosis levels than Pasteur and Danish BCG strains in both HD and UV groups (p<0.05). In addition, we rule out any confounding factor related to storage and viability of the BCG strains used, since all partitions showed virtually the same pattern: 84%, 96% and 95% for live BCG Moreau, Pasteur and Danish, respectively. Remarkably, all BCG strains also induced higher levels (p<0.05) of IL-18, IL-23 and TGF-β1 in the UV group only. Conclusion: The Moreau BCG strain showed superior ability to induce apoptosis when compared with Pasteur and Danish BCG strains. On the other hand, all BCG strains induced similar patterns of in vitro pro and anti-inflammatory cytokines. The vast majority of the world’s current population has been vaccinated with BCG, with the possible requirement for a booster immunization in adulthood for TB protection. A booster immunization with BCG may protect the immunized individuals and further studies that are currently in progress are needed to better evaluate these findings.
