- W4238962323 abstract "This chapter highlights ferredoxins (Fds) isolated from sulfate-reducing bacteria (SRB). It also describes types and distribution of Fds , purification methods, cluster-binding motifs, electronic and magnetic properties of the iron–sulfur clusters involved, cluster interconversions, and the physiological role of Fds . Fds are simple iron–sulfur proteins that contain prosthetic groups composed of iron and sulfur atoms and play a functional role in electron-transfer processes relevant for SRB metabolism. The study of SRB Fds enables the survey of the different properties of simple iron–sulfur proteins including electron transfer, flexibility in coordination chemistry, and ability to undergo cluster interconversions. The isolation of Fds is largely based on the acidic characteristic of these proteins. Because of their low molecular mass, they can be easily separated from other, higher molecular mass components by gel filtration. Purification is carried out at 4〫 and the buffers should be flushed with an inert gas because some of these proteins are oxygen unstable. Four distinct types of Fds are found in SRB-containing [3Fe-4S], [4Fe-4S], [3Fe-4S] plus [4Fe-4S], and 2 × [4Fe-4S] clusters." @default.
