- W4239702912 abstract "Biodegradable polymeric nanoparticles (NPs) are safe, biocompatible nanosystems with tunable properties and controllable activity. These properties make them suitable candidates for a wide range of applications in the medical field, including drug delivery systems (DDS). Many different polymers, of natural and synthetic origin, are used for the formulation of NPs, with all of them sharing a unique characteristic, their biodegradability. Nanoparticulate systems can be prepared by utilizing different methods depending on the characteristics of the polymer and active ingredient, the site of action, and desirable drug release kinetics. Another important feature is their ability for site-specific drug delivery through passive or active targeting. The many advantages presented from biodegradable nanomaterials, coupled with their long history of medical use (as sutures, etc.) led to the development of a number of DDS. Unfortunately despite the huge research efforts and investment by academia and pharmaceutical companies, success rates of approving novel nanomedicine products have not exceeded 10%, mostly because of failures in terms of efficacy and safety profiles during nonclinical and clinical studies. In conclusion it can be safely said that despite the great advancements exhibited in current research, there still exist several challenges and obstacles to be addressed and overcome, including mainly in vitro/in vivo correlation studies as well as regulatory aspects regarding the safety profile of these systems." @default.
