- W4240718310 abstract "β-amyloid [Aβ] and tau pathology become increasingly prevalent with age; however, the spatial relationship between the two pathologies remains unknown. In vivo amyloid (PiB) and tau (AV-1451) PET enable examination of regional pathological associations in cognitively normal elderly potentially in preclinical Alzheimer’s disease. We examined cross-sectional inter-regional PiB and AV-1451 association patterns, with the aim of characterizing regional specificity in PiB—AV-1451 relations. 43 cognitively normal older adults (78.70 ± 5.2 y, 30 female) received 18F-AV-1451 (80-100 min SUVR, cerebellar grey ref.) and 11C-PiB (35-90 min DVR, cerebellar grey ref.) PET and 1.5T magnetic resonance imaging (MRI; 80.58 ± 112.6 d AV-1451—PiB difference; 98.95 ± 119.7 d AV-1451—MRI difference), with images transformed to a common space. We calculated each subject’s mean AV-1451 and PiB uptake values within 87 regions of interest (ROI), and a global cortical PiB value. We examined all pairwise ROI PiB—AV-1451 partial correlations for strength and significance to identify inter-tracer, inter-regional association patterns (covaried for age, sex, and global gray matter volume). Pairwise ROI analysis demonstrated positive and negative PiB—AV-1451 associations (Figure 1). Importantly, strong (p < .01) positive partial correlations were identified between temporal AV-1451 and PIB in extensive temporal and extra-temporal cortical ROIs. We also found less frequent positive associations of regional PiB with frontoparietal AV-1451 (Figure 2). For AV-1451 ROIs where uptake was strongly predicted by PiB ROI (e.g., temporal, frontal AV-1451 uptake), these strong correlations were present regardless of PiB ROI location. Overall correlation strengths were visualized on a brain template (Figure 3). PiB—AV-1451 inter-regional partial correlation (r) values. Aβ and tau pathology, measured using PiB and AV-1451 PET, show significant associations among cognitively normal elderly. In particular, increased PiB uptake both within and outside temporal lobes correlates with increased temporal lobe AV-1451 uptake. In addition, the correlations between Aβ and tau accumulation do not appear to be explained by the location of Aβ accumulation. These results suggest a regional vulnerability of certain brain regions (e.g. temporal lobe) to tau accumulation regardless of where Aβ accumulates." @default.
