- W4240749871 abstract "Extensive amyloid deposition using PET with Pittsburgh Compound B (PiB) has been reported in a large number of normal older subjects who have no clinical impairment. We piloted a recently developed test of controlled associative learning called the Memory Capacity Test (MCT, H. Buschke) to determine if a more challenging memory test would be sensitive to brain amyloid deposition in normal older adults in contrast to traditional memory tests such as the Selective Reminding Test (SRT) and Free and Cued Selective Reminding Tests (FCSRT). Thirty subjects with Clinical Dementia Rating Scores = 0 (mean age = 72.2 ± 8.4, education = 16.4±2.8; AMNART IQ = 124.4±5.5; MMSE = 29.2±0.8, and Geriatric Depression Scale (GDS) = 3.2±3.6) underwent PiB PET and neuropsychological assessment. Mean cortical PiB distribution volume ratio (DVR) was calculated, and a threshold was applied to classify each subject as positive (N = 18) or negative (N = 12). Positive and negative groups did not differ on mean age, MMSE, education, AMNART IQ or GDS. However, the PiB-positive subjects performed less well on several subtests of the SRT including Long Term Store (p = 0.05), Long Term Retrieval (p = 0.04), Continuous Retrieval (p = 0.04), and Delayed Recall (p = 0.03) but not Total Recall (Mann-Whitney U). The MCT was significantly lower in PiB-positive subjects for 2nd list learning (p = 0.03) in contrast to 1st list learning, and 30-minute delayed recall of the 2nd list (p = 0.002). No significant differences were found on the FCSRT for either Free or Cued Recall. Our findings suggest that more challenging tests of memory, those involving either selective reminding procedures or increased associative list learning, may provide greater sensitivity for detecting subtle memory impairment mediated by amyloid deposition among normal older individuals, compared to standard memory tests used for detecting impairment in patients with Alzheimer's disease. These findings suggest that more challenging memory tests may be needed for anti-amyloid treatment trials in preclinical populations." @default.
