Learn more.Copy URL Share a linkShare onEmailFacebookTwitterLinked InRedditWechat Abstract Front cover: Increasing evidence suggest that the secretome would be an ideal alternative of mesenchymal stem cells (MSCs) transplantation in the treatment of traumatic brain injury (TBI). Transplanted MSCs can sense and response to regeneration signals in the injury microenvironment after TBI. Therefore, it is reasonable to hypothesis that signaling molecules secreted in response to local tissue damage can further promote the therapeutic effect of MSCs secretome. To most closely replicate the complex microenvironment in injured brain, we used extract of traumatic brain tissue to preconditioning umbilical cord mesenchymal stem cells (UCMSC) in vitro, and stereotaxically injected these secretome into dentate gyrus of hippocampus in a rat severe TBI model. Sox2 is a major mediator of Notch signaling in maintaining the precursor pool in the adult subgranular zone. The administration of the preconditioned UCMSC secretome with injured brain extract further increased the level of Sox2 compared with that treated with normal brain extracts. We observed that the injury-preconditioned UCMSC secretome could significantly further enhanced cell proliferation in the dentate gyrus after traumatic brain injury. Our study utilized the injury-preconditioning secretome to inducibly amplify the neurogenesis and cognitive recovery, which can offers a novel and safer candidate for nerve repair.
