- W4250142064 abstract "Successful species conservation requires the maintenance of a sustainable population with sufficient genetic diversity. Reproductive sciences have the potential to greatly support efforts to conserve rare and threatened species. The generation of induced pluripotent stem cells from adult somatic cells has been extensively investigated for various research and therapeutic applications, but they may also play an important role in species conservation. Currently, tissues and somatic cell cultures from numerous species have been cryopreserved for future use in biobanks across the world representing a valuable source of genetic material that can be immortalized through the generation of iPSCs. This material can provide a limitless supply of cells for various applications ranging from fundamental research to the generation of live offspring. The production of embryos by tetraploid complementation or production of competent oocytes and spermatozoa through controlled differentiation processes would finally allow in vitro fertilization, in vitro embryo culture (IVC), and embryo cryopreservation and transfer to become accessible on a large scale. The concept of “in vitro breeding,” the notion of creating embryos from oocytes and sperm generated completely in vitro, would also become a reality. While iPSCs can exert a significant impact on how reproductive sciences support species conservation, much research needs to be done before these possibilities can materialize. Many challenges exist, including the importance of maintaining the purity of the threatened species genome, and addressing species-specific differences in basic cell culture requirements and iPSC reprogramming protocols. This chapter discusses the particular advantages of iPSC technology for species conservation, as well as its various potential applications, including the generation of iPSC-derived gametes and live offspring." @default.
