The blot was derived from the same Akt1 silencing experiment with two independent Akt1 siRNA sequences. In Figure 1D, cells were assayed for Matrigel invasion, and, in Figure 3B, a fraction of the same transfectants was assayed for NFAT IL-2 luciferase activity. Because the two sets of data were derived from the same experiment, we showed the same anti-Akt1 immunoblot, but this was not stated in the legend to either figure. We now publish an independent experiment equivalent to the original Figure 3B. The two Akt1 siRNA sequences were transiently transfected into SUM-159-PT cells along with the NFAT IL-2-luciferase reporter plasmid and β-galactosidase. Again, this shows increased NFAT transcriptional activity in cells in which Akt1 expression has been knocked down, along with corresponding Akt1 and actin immunoblotting. Error bars indicate ±SD.
