However, successful gene therapy depends on the delivery of the therapeutic genes into appropriate target cells. As stem cells can be relatively easily transduced with viral vectors, the durable expression of therapeutic genes is expected. This study was performed to examine the capability of HGF expressing human mesenchymal stem cells(hMSCs) to inhibit collagen deposition in bladder outlet obstruction(BOO) of rats. METHODS Immortalized human mesenchymal stem cells(hMSCs) lines with a retroviral vector encoding v-myc was prepared. HGF was transfected with recombinant retrovirus from the pLPCX.HGF. Six week old female Sprague-Dawley rats were divided into 4 groups of 10 each, including group 1-sham operation, group 2- BOO for 4 weeks, group 3- hMSCs treatment and group 4- HGF expressing hMSCs treatment after BOO. Two weeks after the onset of BOO, hMSCs was injected into the bladder wall in group 3, HGF expressing hMSCs was injected into the bladder wall in group 4. After 4 weeks, bladder tissues were harvested and Masson′s trichrome staining was performed. Each slide was inspected microscopically and the mean percent collagen area was examined according to the formula, collagen/collagen + muscle. Intraperitoneal cyclosporine with 10mg/kg injection was done everyday in group3 and 4. RESULTS Transgene expression in HGF expressing hMSCs was demonstrated by reverse transcriptase polymerase chain reaction. Enzymatic activity of HGF expressing hMSCs was confirmed by enzyme linked immunosorbent assay. The mean bladder weight was 0.11±0.06g, 0.72±0.35g, 0.67±0.25g, 0.27±0.09g in group 1 to 4 respectively. Group 2 showed higher bladder weight than group 1(p<0.05). Group 3 showed lower bladder weight than group 2 and Group 4 showed lower bladder weight than group 3(p<0.05, respectively). The mean percent of collagen area was 12.2±1.5%, 27.9±2.8%, 25.4±2.7%, 15.2±0.8% in group 1 to 4 respectively. Group 2 showed higher mean percent of collagen area than group 1(p<0.05). Group 3 showed lower mean percent of collagen area than group 2 and Group 4 showed lower mean percent of collagen area than group 3(p<0.05, respectively). CONCLUSIONS HGF expressing hMSCs inhibited collagen deposition in BOO of rats. This finding may imply a possible novel therapeutic strategy against bladder fibrosis in patients with BOO.
