- W4254709984 abstract "This chapter focuses on a number of criteria for setting up a super-resolution microscopy system. It provides a discussion of major hardware components, such as the optical elements for collection of fluorescence signals, detectors, modes of illumination, adaptive optics, the computer technology, as well as a conceptualization of their interplay in the overall system. Furthermore, the software requirements are analyzed: feature extraction, error correction, visualization, meta-analysis and confidence analysis. As one of the most important parts of a super-resolution microscope, the objective lens and its characteristics are reviewed, because it defines light collection efficiency and quality of the imaged signals. Various array detectors are described with respect to their characteristic sensitivity, noise, and frame rates. In conventional fluorescence microscopy, the images can be displayed and used in principle without further processing. In contrast, the software used for image analysis and visualization has a strong impact on super-resolution imaging. Major sources of aberrations are covered, including approaches to correct for deteriorations in the images, as implemented e.g. in adaptive optics. The chapter discusses the procedures for feature extraction (e.g. by fitting a model function), necessary calibration and correction steps for quantitative imaging, approaches to obtain reconstructions and visualizations of the observed structures, and particulars of feature extraction in structured illumination microscopy. It gives a short overview of meta-analyses used to study e.g. local structure density, clustering, or morphological parameters, which do not rely on image visualizations. It concludes with a discussion of approaches to verify or to challenge the reliability of the extracted or reconstructed data, as well as of open source and best practice initiatives in the realm of super-resolution microscopy." @default.
