- W4280492288 abstract "Antiplatelet therapy represents a major pharmacological strategy aimed at reducing the risk of atherothrombosis-associated cardiovascular events. In this chapter, the authors review the evidence supporting the importance of heme oxygenase (HO)-derived carbon monoxide (CO) in the regulation of thromboresistance in vivo , in particular exemplified by CO-dependent inhibition of platelet function. They also review emerging evidence showing that CO affords antiplatelet activities that could be mechanistically explained by the effects of CO in modulating intraplatelet energy metabolism resulting in NAD + and adenosine triphosphate deficiency. These very recent findings might open a new avenue for antiplatelet strategies based on inhibition of platelet energy metabolism. The first attempts to assess the effects of exogenous CO on platelet functions were undertaken in the 1970s to 1980s, still prior to the discovery of intracellular CO-generating enzymes, namely HO-1 and HO-2." @default.
