- W4283172015 abstract "TYPE: Late Breaking Abstract TOPIC: Lung Cancer PURPOSE: To Establish an effective molecular differential diagnosis tool for primary lung cancer (MP) and intrapulmonary metastasis (IM). METHODS: We performed panel-based and whole-exomes sequencing on 31 tumor and 10 blood samples from 10 patients with MP or IM. RESULTS: Under panel-sequencing assessment, seven patients were confirmed as MP while three patients were assessed as IM (Figure 1A). The panel-sequencing results were later confirmed by WES-based characterization (Figrue 1B). Based on the molecular diagnosis, we used the lasso algorithm to identifiy candidate genes for classifiers (Figrue 1C). We found that the discriminatory abilities of panel sequencing and WES classifier were similar (Figrue 1D). CONCLUSIONS: Our results proved that establishment of molecular classifiers with an optimised number of genes for multifocal lung cancer is possible. However, further analysis with larger sample size is needed to establish a more robust and universal panel-based diagnostic tool for these lung cancer entities. CLINICAL IMPLICATIONS: This study shows that clinical panel sequencing and WES can effectively distinguish between primary lung cancers and intrapulmonary metastases. Furthermore, the findings in this study suggest a possibility that more efficient genetic models with lesser genes will be developed in the future with a support of genomic data from more clinical samples.
