- W4286255342 abstract "CXCL20a is a teleost-specific chemokine with antimicrobial peptide-like and bacterial aggregation functions. It is composed of an extended N-terminal coil, a three-stranded antiparallel β-sheet, an α-helix, and a C-terminal coil. However, which specific structure plays a key role in the antibacterial activity? In this study, we found that all the four truncated peptides from CiCXCL20a on the basis of its secondary structure exert potent antimicrobial activity against Gram-negative (G−) and Gram-positive (G+) bacteria with negligible toxicity. All the CiCXCL20a truncated peptides can kill bacteria, disrupt the integrity of bacterial cell membranes and result in bacterial cell cytoplasm leakage. We further identified that CiCXCL20a aggregates Gram-negative bacteria mainly through CiCXCL20a C-terminal coil region. In the grass carp (Ctenopharyngodon idella) model infected with a lethal dose of Aeromonas hydrophila, all the CiCXCL20a truncated peptides can significantly reduce tissue bacterial loads and mortalities. In addition, we also confirmed that all the CiCXCL20a truncated peptides can significantly suppress A. hydrophila-stimulated inflammation in vivo by suppressing the production of the pro-inflammatory cytokine CiTNF-α and increasing the production of the anti-inflammatory cytokine CiIL-10. The present study adds further evidence for the close functional and evolutionary relationships between chemokines and antimicrobial peptides. Moreover, the effective antimicrobial and anti-inflammatory activities of all the CiCXCL20a truncated peptides in the teleost model suggest that all the CiCXCL20a truncated peptides can be promising novel therapeutic agents for bacterial diseases and inflammatory disorders." @default.
