- W4286598082 abstract "Cellular senescence is a debatably irreversible block on the proliferation of potentially oncogenic or damaged cells. Senescent cells comprise heterogeneous and evolving states with various phenotypes based on cell type, senescence etiology, and degree of senescence. The accumulation of senescent cells with age and disease progression reflects a combination of dysfunctional senescent cell clearance and upregulated senescence induction by stress, telomere erosion, paracrine signaling, and DNA damage. A hallmark of senescent cells is the secretion of proinflammatory proteins, and growth and necrotic factors, in an expression phenotype termed the senescence-associated secretory phenotype (SASP). SASPs modulate the immune response to senescent cells and can induce senescence in other cells. This creates a positive feedback loop of induction and inflammation, which may explain its association with chronic inflammatory disorders such as Alzheimer’s, diabetes, and chronic kidney disease. In this chapter, we discuss the potency of senescent cell-targeting therapeutics, or “Senotherapeutics,” as strategies against age-related disorders. Clearing senescent cells improves cell viability in vitro and tissue function, halts the progression of age-related disorders, and increases lifespan in vivo. We explore “Senolytics,” molecules that induce cell death in senescent cells, immunomodulators that condition the immune response, vaccines that may boost immune cell-mediated clearance of senescent cells, senescence-reversing agents, and SASP-inhibitory agents that aid net clearance by preventing further senescence induction. As senotherapeutics begin to populate clinical trials, we outline the challenges in drug repurposing and efficacy, and target identification that will decide their future in treating age-related disorders." @default.
