- W4292395538 abstract "The clinical utility of immune checkpoint blockade has profoundly changed the treatment landscape and outlook for patients in a large number of cancer types, amounting to long-term survival benefits in patients for whom there were limited options to do so just a decade ago. Despite this breakthrough addition to the arsenal of therapeutic options available to treating physicians, both primary and secondary resistance to immune checkpoint blockade occurs in a substantial number of patients. The expansion of clinical use of checkpoint blockade both as standard of care and in clinical trials has lead to the accumulation of clinical biomarker data from patients that do or do not respond to therapy, providing an opportunity to identify particular patient profiles or characteristics associated with resistance to checkpoint inhibition. While these biomarker data serve as a critical means with which to generate translationally relevant hypotheses around mechanisms of resistance, appropriate preclinical models that can recapitulate the disease biology in particular patient subsets are required in order to determine whether there is a causative or mechanistic link between the observed biomarker data and patient outcomes on treatment. This chapter aims to highlight the role that preclinical mouse models play as critical tools in the effort to understand both the molecular and cellular mechanisms of resistance to checkpoint blockade and also to identify rationale combination approaches to deliver therapeutic options to cancer patients with high unmet need, including those with clinical resistance to checkpoint blockade." @default.
