- W4296617492 abstract "Nine tetracycline antibiotics are currently on the market. Only one fully synthetic tetracycline has been approved by the FDA. That is Tetraphase Pharmaceuticals' hyper-potent eravacycline. Tetracyclines became the most prescribed broad-spectrum antibiotics in the United States within three years. Overcoming drug resistance requires constant replenish our arsenal of antibiotics. Ribosomal protection proteins are cytoplasmic proteins that bind to the ribosome, causing an alteration in ribosomal conformation that prevents tetracycline from binding. Tetracycline antibiotics are quite fragile. They decompose readily at pH 10 or higher, which explains why limited semi-synthetic tetracyclines are available on the market. The first- and the second-generation syntheses of eravacycline were developed by Myers' group at Harvard. Myers' convergent and practical synthesis of eravacycline demonstrated the power of total synthesis, which led to access to many tetracyclines that were not possible from semi-synthesis." @default.
