The transition of short-term memories to long-lasting memories are environmental stimuli-driven, and this depends on several underlying mechanisms in the central nervous system regulated by modifications in transcriptional and translational pathways, downstream signaling pathways, pre- and postsynaptic receptor activation, protein synthesis, and gene expression regulation. Studies have identified the critical role of posttranslational modifications of histones and covalent modifications on DNA as major regulators of memory processes. Their dynamic activity resulted in long-term memory consolidation in different types of memory processes followed by neuronal activation. Further, pharmacological manipulations of these modifications have been studied to understand the molecular mechanisms involved and also to exploit these modifications therapeutically in treating several neurodegenerative disorders or diseases. Recently, new regulators of memory were identified, known as histone variants, which by exchanging with canonical histones promote transcription and thus are involved in the memory consolidation processes. Epitranscriptomics, exploiting the role of mRNA modifications, have also been studied recently in relation to long-term memory formation. This book chapter details the cellular and molecular mechanisms that govern the gene expression states and their role in learning and memory by reviewing the evidences reported so far and also by citing recent advances in the field of neuroepigenetics, thus providing an incite for research in the future." @default.
