- W4306177763 abstract "INTRODUCTION: Mesenchymal stem cells provide protection during experimental necrotizing enterocolitis (NEC) through the release of hydrogen sulfide (H2S). Overexposure to H2S can be toxic, and previous methods of H2S measurement have been ineffective. We set out to synthesize a near-infrared H2S (NIR-H2S) probe and hypothesized that the probe would allow for qualitative assessment of H2S in biologic systems. METHODS: NIR-H2S was synthesized using a method described by Zhang et al (Scientific Reports 2016). HIEC6 cells were treated with NaHS (H2S donor) or ZnCl2 (negative control) in the presence of NIR-H2S probe. In separate experiments, lentivirus vectors designed to overproduce enzymes involved in endogenous H2S production (cystathione-β-synthase [CBS], cystathione-γ-lyase [CTH], and mercaptopyruvate sulfur transferase [MPST]) were used to infect HEK293T cells and umbilical cord–derived mesenchymal stem cells (USCs). Adult C57Bl6 mice were injected intraperitoneally with NaHS in the presence of the probe. Mice were imaged with IVUS Spectrum CT. RESULTS: H2S was imaged in HIEC6 cells and mice using NIR-H2S probe (Figs. 1A–1C). Probe activity was increased in the presence of NaHS. HEK293T cells infected with lentivirus containing CBS, CTH, or MPST showed overexpression of these enzymes by Western blotting analysis (Figs. 1D–1F). Stable transfection and selection of lentivirus infected USCs was confirmed by expression of the reporter gene GFP (Figs. 1F).CONCLUSION: H2S production was successfully imaged in both in vitro and in vivo systems. The ability to assess H2S production from stem cells during therapeutic use will allow for the safe application of these cells during therapy for NEC." @default.
