- W4308366418 abstract "シヌクレイノパチーはパーキンソン病（PD）やびまん性レビー小体病（DLBD）や多系統萎縮症（MSA）などを含む．140アミノ酸からなるαシヌクレイン（αSyn）の異常な蓄積によって起こる神経変性疾患である．PD及びDLBDでは，神経細胞においてαSynが誤って折りたたまれ，凝集してレビー小体（Lewy body：LB）とレビー神経突起（Lewy neurite：LN）を形成する．一方，MSAでは，αSynは主にオリゴデンドロサイト（OLG）に蓄積し，グリア細胞質封入体（GCI）を形成する．LB，LN，GCIのαSynの凝集体はシヌクレイノパチーを特徴づける病理である．同時に，細胞死を誘発する原因である．しかし，細胞死を誘発するメカニズムは不明である．私達は，脂肪酸結合タンパク質（FABP）がαSynと毒性のある複合体（ヘテロオリゴマー）を形成し，シヌクレイノパチーの細胞死に関与することを見出した．脂肪酸結合タンパク質3（FABP3）はPDのドパミン神経細胞死に，脂肪酸結合タンパク質7（FABP7）はOLGの細胞死に関与する．本稿では，MSAにおけるαSynとFABP7の役割とFABP7を標的にした新規治療薬開発について報告する．" @default.
