- W4308592094 abstract "Adult neural stem cells (NSCs) located in defined regions of the mammalian brain continuously produce new neurons throughout life. This process of stem cell-driven neurogenesis is considered as a form of structural plasticity that allows the mature brain to adapt to its environment. Preservation of a healthy proteome through autophagy is key for maintaining NSC function. The autophagy lysosomal pathway is directly linked to the activation of quiescent NSC reservoirs and has a cell-autonomous role in the maturation of newborn neurons. However, excessive autophagy can also lead to autophagic cell death of the adult NSCs. Since NSC homeostasis and neurogenesis decline during ageing and are altered in several pathological conditions, autophagy is emerging as a relevant pharmacological target.
