During epithelial polarisation of embryonic stem cells, the site of apical membrane initiation is defined by E‐cadherin mediated cell adhesion independently of cell division. Image shows chimeric cell doublets and epithelial cysts labelled with E‐cadherin in Magenta, F‐actin in blue and PAR‐3 in green. Only doublets with E‐cadherin in both cells form a PAR‐3 positive AMIS. Whilst epithelial cysts expressing E‐cadherin formed lumens via hollowing, those without E‐cadherin formed lumens via folding.
