- W4309356005 abstract "Drug metabolite kinetics studies the formation and disposition kinetics of metabolites after drug administration. This chapter reviews the concepts and principles of in vivo drug metabolite kinetics along with examples. All doses, drug amounts, and drug concentrations are in molar units to avoid conversions between a drug and a metabolite due to the difference in molecular weight. In a drug–drug interaction study, metabolite concentrations associated with an affected clearance pathway are sometimes measured. The elimination clearance of a metabolite can be determined by intravenous dosing of a metabolite synthetic standard, with a caveat that disposition kinetics may be different between an in situ -generated metabolite and an exogenously dosed one. Pharmacokinetic/pharmacodynamic (PK/PD) modeling is a useful tool to evaluate the quantitative relationship between drug exposure and pharmacological response. Several approaches can be considered to incorporate pharmacologically active metabolites into the PK/PD modeling." @default.
