- W4310367132 abstract "Pyrin is an inflammasome sensor that acts as a guard of RhoA GTPases. It is activated in response to numerous bacterial pathogens secreting toxins or effectors that inhibit RhoA. In addition, the Pyrin inflammasome is implicated in several monogenic autoinflammatory diseases. Particularly, mutations in MEFV gene, which encodes Pyrin, can cause two distinct diseases called familial Mediterranean fever (FMF) and Pyrin-associated autoinflammation with neutrophilic dermatosis (PAAND). Here, we review the molecular mechanisms of Pyrin activation following RhoA inhibition, highlighting the key role of the dephosphorylation and of the cytoskeleton. We will then present its regulation by both transcriptional and posttranslational mechanisms and the physiological and pathophysiological consequences of Pyrin inflammasome activation during infections and autoinflammatory diseases." @default.
