- W4310367147 abstract "The NLR family, apoptosis inhibitory protein (NAIP)/NLR family CARD domain-containing protein 4 (NLRC4) is the most studied inflammasome complex in the context of host defense against infections. NAIP members function as sensors for bacterial ligands such as flagellin, “needle,” and “rod” proteins, whereas NLRC4 serves as a platform to recruit caspase-1 or the adapter molecular ASC (apoptosis-associated speck-like protein containing a caspase activating and recruitment domain). NAIP/NLRC4 activation results in a series of effector mechanisms to control infections including pyroptosis, IL-1β and IL-18 secretion, eicosanoid storm, intestinal epithelial cells (IEC) expulsion, and epigenetic regulation of inducible Nitric Oxide Synthase 2 (iNOS/NOS2). Recent evidence has been demonstrating the role of NAIP/NLRC4 beyond bacterial infections. NAIP/NLRC4 seems to participate in fungal, protozoan, and viral infections, and, also in sterile inflammatory pathologies, although the molecular mechanisms involved in inflammasome activation in these situations remain to be elucidated. Here we will discuss recent advances in the NAIP/NLRC4 biology and its relevance to the host resistance against infections and inflammatory pathologies." @default.
