Furthermore, mitochondria are actively involved in the intrinsic apoptotic pathway as well as in necrosis and in autophagy. Because mitochondria are involved both in vital and in lethal functions in physiological and pathological settings, their role in cancer initiation and progression has been extensively studied. Many anticancer drugs exert their cytotoxic activity both directly and indirectly on mitochondria. However, the other face of Janus is that the mitochondrion itself, also of non-tumoral cells, is the target of many adverse effects of numerous chemotherapeutic drugs. Here, we explored the case of three different chemotherapeutics (doxorubicin, trastuzumab, and sunitinib) widely used in clinical practice and whose main side effect is cardiotoxicity linked to their ability to damage the mitochondria of cardiomyocytes." @default.
