Over the decades, microfluidics has boosted the development of single-cell RNA sequencing (scRNA-seq) techniques and gradually been applied in spatial transcriptomics. Microfluidics with the merits of miniaturization, integration, and automation provide mainstream scRNA-seq platforms to interrogate cellular heterogeneity and multimodal biological information with high sensitivity, excellent accuracy, and low cost. Microfluidics also applies to encode tissue with high spatial resolution and allows spatial multi-omics analysis. Herein, microfluidics-assisted transcriptomics and multi-omics methodologies from single-cell and spatiotemporal dimensions are reviewed. First, this review focuses on single-cell transcriptomics technologies concerning high throughput, high cell utilization, and rare cell analysis. Second, microfluidics-based single-cell multi-omics studies are summarized. The fast-growing microfluidics-assisted temporal transcriptomics, spatial transcriptomics, and spatial multi-omics approaches are then highlighted. Finally, the advantages, challenges, and future developments of these approaches are discussed." @default.
