• W4312367046 abstract "<b>Introduction:</b> The role of <i>Neisseria</i> species in bronchiectasis is unknown. <b>Methods:</b> Bacteriome sequencing of airway samples from the Cohort of Asian and Matched European Bronchiectasis (CAMEB) (n=225) was performed to assess clinical correlates of disease associated with <i>Neisseria spp</i>. Mouse lung infection and primary human epithelial cell models were employed to assess the pathogenic potential of <i>Neisseria</i>. Transcriptomic, metabolomic and lipidomic analyses identified systemic changes in mice following <i>Neisseria</i> exposure, which was validated in human subjects by serum metabolipidomic analysis of <i>Neisseria</i>-dominant patients. Metagenomic surveillance of airway and environmental samples was employed to assess the broader implications of the <i>Neisseriaceae</i> in chronic respiratory disease. <b>Results:</b><i>Neisseria</i>-dominant microbiomes associate with poorer clinical outcomes. <i>N. subflava</i> promotes loss of epithelial integrity and inflammation in primary epithelial cells while in-vivo animal models exhibit immuno-inflammatory functional gene clusters and pulmonary inflammation. Findings from <i>Neisseria</i>-infected mouse metabolipidomes were validated in human <i>Neisseria</i>-dominant bronchiectasis, corroborating its potential pathobiont role. Metagenomic surveillance identifies <i>Neisseria</i> species as a pathobiont across other chronic respiratory diseases and confirms detection in the home environment of bronchiectasis patients. <b>Conclusion:</b><i>Neisseria</i> species represent novel pathobionts in bronchiectasis. <b>Funding:</b> Singapore Ministry of Health’s National Medical Research Council under its Clinician-Scientist Individual Research Grant (MOH-000141) (S.H.C) and Clinician Scientist Award (MOH-000710) (S.H.C)." @default.
