Neoplastic cells produce elevated levels of reactive oxygen species that trigger signaling pathways and specific gene expression to aid cancer cell proliferation and activate stress responsive, adaptive cellular metabolic programs. Among other steps, the final stage of gene expression comprising of protein synthesis is extensively and dynamically modulated in response to stress and in cancer. This chapter first outlines the process of mRNA translation and its regulation followed by how this is reprogramed in cellular stress and during oncogenesis, especially in the context of oxidative stress. Targeting mRNA translation for cancer therapeutics is an emerging field that has enhanced possibilities for combination therapy. Owing to the intimate links between oxidative stress and remodeling of cellular mRNA translation, the chapter concludes by emphasizing the necessity to investigate outcomes of combining standard of care anti-oxidants/ROS enhancers with translation inhibitors in treatment of difficult to cure cancers." @default.
