- W4312547946 abstract "<b>Introduction:</b> In the normal conducting airway, the reticular basement membrane (RBM) is composed of extracellular matrix (ECM) proteins laminin and collagen IV. In the asthmatic lung, the RBM additionally contains collagen I, III and fibronectin. <b>Objective:</b> Assess the effect of different RBM ECM proteins on airway epithelial cell attachment and barrier formation using real-time electrical cell substrate impedance sensing (ECIS). <b>Methods:</b> ECIS arrays were coated with fibronectin, collagen I, III, IV, laminin, and bovine serum albumin as control. The airway epithelial cell line (1HAEo-) was seeded and monitored for 80h to assess cell attachment, spreading and barrier formation. Data were analyzed using a generalized additive mixed effect model (GAMM) and one-phase decay models. <b>Results:</b>1HAEo-cells attached (cover electrode area/hr) and spread the fastest (cell-spreading rate/hr) when grown on fibronectin, followed by collagen I and III compared to collagen IV, laminin and control. Complete epithelial barrier formation of 1HAEo-cells was the shortest when grown on fibronectin and collagen I, then collagen III and IV, compared to collagen IV, laminin and control. Paracellular junction formation began at 8h for fibronectin and collagen I, by 10h for collagen III and IV, and 20h for laminin and control. Lastly, fibronectin and collagen I enabled the closest attachment of 1HAEo-cells via basal adhesions. <b>Conclusions:</b> Airway epithelial cells attach, spread, and form a barrier fastest on fibronectin and collagen I. These ECM proteins may preserve the epithelial barrier during airway remodeling in asthma." @default.
